Nitrifying and denitrifying bacteria in aerobic granules formed in sequencing batch airlift reactors

摘要: The purpose of this study was to investigate nitrifying bacteria and denitrifying bacteria isolated from aerobic granules. Aerobic granules were formed in an internal-circulate sequencing batch airlift reactor (SBAR) and biodegradation of NH3--N was analyzed in the reactor. Bacteria were isolated and determined from aerobic granules using selected media. The growth properties and morphology of bacteria colonies were observed by controlling aerobic or anaerobic conditions in the culture medium. It was found that bacteria in aerobic granules were diverse and some of them were facultative aerobes. The diversity of bacteria in aerobic granules was a premise of simultaneous nitrification and denitrification.

摘要: To investigate the nitrogen transport and conversion inside activated sludge flocs, micro-profiles of O , NH , NO , NO , and pH were measured under different operating conditions. The flocs were obtained from a laboratory-scale sequencing batch reactor. Nitrification, as observed from interfacial ammonium and nitrate fluxes, was higher at pH 8.5, than at pH 6.5 and 7.5. At pH 8.5, heterotrophic bacteria used less oxygen than nitrifying bacteria, whereas at lower pH heterotrophic activity dominated. When the ratio of C to N was decreased from 20 to 10, the ammonium uptake increased. When dissolved oxygen (DO) concentration in the bulk liquid was decreased from 4 to 2 mg·L , nitrification decreased, and only 25% of the DO influx into the flocs was used for nitrification. This study indicated that nitrifying bacteria became more competitive at a higher DO concentration, a higher pH value (approximately 8.5) and a lower C/N.

Microprofiles of activated sludge aggregates using microelectrodes in completely autotrophic nitrogen removal over nitrite (CANON) reactor

摘要: Microsensor measurements and fluorescence in situ hybridization (FISH) analysis were combined to investigate the microbial populations and activities in a laboratory-scale sequencing batch reactor (SBR) for completely autotrophic nitrogen removal over nitrite (CANON). Fed with synthetic wastewater rich in ammonia, the SBR removed 82.5±5.4% of influent nitrogen and a maximum nitrogen-removal rate of 0.52 kgN·m ·d was achieved. The FISH analysis revealed that aerobic ammonium-oxidizing bacteria (AerAOB) and anaerobic ammonium-oxidizing bacteria (AnAOB) dominated the community. To quantify the microbial activities inside the sludge aggregates, microprofiles were measured using pH, dissolved oxygen (DO), , and microelectrodes. In the outer layer of sludge aggregates (0–700 μm), nitrite-oxidizing bacteria (NOB) showed high activity with 4.1 μmol·cm ·h of maximum nitrate production rate under the condition of DO concentration higher than 3.3 mg·L . Maximum AerAOB activity was detected in the middle layer (depths around 1700 μm) where DO concentration was 1.1 mg·L . In the inner layer (2200–3500 μm), where DO concentration was below 0.9 mg·L , AnAOB activity was detected. We thus showed that information obtained from microscopic views can be helpful in optimizing the SBR performance.

Cultivation of aerobic granular sludge in a conventional, continuous flow, completely mixed activated

摘要: Aerobic granules were formed in a conventional, continuous flow, completely mixed activated sludge system (CMAS). The reactor was inoculated with seed sludge containing few filaments and fed with synthetic municipal wastewater. The settling time of the sludge and the average dissolved oxygen (DO) of the reactor were 2 h and 4.2 mg·L , respectively. The reactor was agitated by a stirrer, with a speed of 250 r·min , to ensure good mixing.The granular sludge had good settleability, and the sludge volume index (SVI) was between 50 and 90 mL·g . The laser particle analyzer showed the diameter of the granules to be between 0.18 and 1.25 mm. A scanning electron microscope (SEM) investigation revealed the predominance of sphere-like and rod-like bacteria, and only few filaments grew in the granules. The microbial community structure of the granules was also analyzed by polymerase chain reaction-denaturing gradient gel electrophoresis (PCR-DGGE). Sequencing analysis indicated the dominant species were α, β, and γ- , , and . The data from the study suggested that aerobic granules could form, if provided with sufficient number of filaments and high shear force. It was also observed that a high height-to-diameter ratio of the reactor and short settling time were not essential for the formation of aerobic granular sludge.

Impact of total organic carbon and chlorine to ammonia ratio on nitrification in a bench-scale drinking water distribution system

摘要: Nitrification occurs in chloraminated drinking water systems and is affected by water quality parameters. The aim of this study was to investigate the impact of total organic carbon and chlorine to ammonia ratio on nitrification potential in a simulated drinking water distribution system as during chloramination. The occurrence of nitrification and activity of nitrifying bacteria was primarily monitored using four rotating annular bioreactors (RAB) with different chlorine to ammonia ratios and total organic carbon (TOC) levels. The results indicated that nitrification occurred despite at a low influent concentration of ammonia, and a high concentration of nitrite nitrogen was detected in the effluent. The study illustrated that reactors 1(R1) and 3 (R3), with higher TOC levels, produced more nitrite nitrogen, which was consistent with the ammonia-oxidizing bacteria (AOB) counts, and was linked to a relatively more rapid decay of chloramines in comparison to their counterparts (R2 and R4). The AOB and HPC counts were correlated during the biofilm formation with the establishment of nitrification. Biofilm AOB abundance was also higher in the high TOC reactors compared with the low TOC reactors. The chlorine to ammonia ratio did not have a significant impact on the occurrence of nitrification. Bulk water with a high TOC level supported the occurrence of nitrification, and AOB development occurred at all examined chlorine to ammonia dose ratios (3∶1 or 5∶1).

Optimization of thermophilic anaerobic-aerobic treatment system for Palm Oil Mill Effluent (POME)

摘要: Optimization of an integrated anaerobic-aerobic bioreactor (IAAB) treatment system for the reduction of organic matter (Chemical Oxygen Demand (COD), Biochemical Oxygen Demand (BOD) and Total Suspended Solids (TSS) concentrations) in Palm Oil Mill Effluent (POME) to legal standards with high methane yield was performed for the first time under thermophilic condition (50°C–55°C) by using response surface methodology (RSM). The experiments were conducted based on a central composite rotatable design (CCRD) with three independent operating variables, organic loading rates in anaerobic compartment (OLR ) and mixed liquor volatile suspended solids (MLVSS) concentration in anaerobic (MLVSS ) and aerobic compartments (MLVSS ). The optimum conditions for the POME treatment were determined as OLR of 15.6 g COD·L ·d , MLVSS of 43100 mg·L , and MLVSS of 18600 mg·L , where high aerobic COD, BOD and TSS removal efficiencies of 96.3%, 97.9%, and 98.5% were achieved with treated BOD of 56 mg·L and TSS of 28 mg·L meeting the discharge standard. This optimization study successfully achieved a reduction of 42% in the BOD concentrations of the final treated effluent at a 48% higher OLR as compared to the previous works. Besides, thermophilic IAAB system scores better feasibility and higher effectiveness as compared to the optimized mesophilic system. This is due to its higher ability to handle high OLR with higher overall treatment efficiencies (more than 99.6%), methane yield (0.31 L CH ·g COD ) and purity of methane (67.5%). Hence, these advantages ascertain the applicability of thermophilic IAAB in the POME treatment or even in other high-strength wastewaters treatment.

Characteristics of night soil and leaf co-composting using aerobic static method

摘要: The main purpose of this work is to investigate the characteristics of night soil and leaf co-composting using aerobic static composting method. Three influencing factors, including proportion of night soil and leaf, aeration rate and aeration pattern, were investigated through the evolution of the principal physicochemical properties, i.e., temperature, oxygen consumption rate, organic matters, moisture content, carbon, nitrogen, carbon-to-nitrogen ratio and Germination index (GI). It was found that the 3∶1 (w∶w) mixture of night soil and leaf was capable of achieving the highest composting temperature, longest retention time of high temperature (55°C), and fastest organic matter degradation. The 0.14 m ·min ·m aeration rate was most beneficial to composting, and the mixture of night soil and leaf maintained the highest temperature for the longest duration and achieved the highest CO content and GI. The continuous aeration pattern during composting was superior to an intermittent aeration pattern, since the latter delayed the composting process.

Aerobic granulation of pure bacterial strain

摘要: The objective of this study is to cultivate aerobic granules by pure bacterial strain, , in a sequencing batch reactor. Stable granules sized 2.0–2.2 mm were formed in the reactor after a five-week cultivation. These granules exhibited excellent settling attributes, and degraded phenol at rates of 1.49 and 1.19 g phenol/(g VSS·d) at 250 and 1500 mg/L of phenol concentration, respectively. Confocal laser scanning microscopic test results show that was distributed over the initial small aggregates, and the outer edge of the granule was away from the core regime in the following stage.
